Chemical treatments were used to produce sex-reversed female (genetic male) Tilapia aurea, which can be bred to produce monosex offspring. Various combinations of estrogens, two synthetic and one natural, an antiandrogen, and a ‘pituitary blocking agent’ were orally administered to sexually undifferentiated fry. Treatment periods were 4, 5, 6 and 8 weeks. Seven of the 36 treatments produced more than 50% females (α = 0.05). Two treatments were more successful than the others: (1) 17-α-ethynylestradiol (EE) at 100 mg kg?1 diet in combination with methallibure (ME) at 100 mg kg?1 for 6 weeks and (2) EE-100 with ME-100 and cyproterone acetate (CA) at 100 mg kg?1 for 6 weeks. These treatments produced fish that were 90% female. Comparisons of the treatments with and without CA indicated that CA may lessen the hormone effectiveness of the treatments. Diethylstilbestrol was as potent as EE in comparable CA treatments.  相似文献

The relationship between photosynthesis and accumulated cold degree days (CDD) over the late growing season was examined at the shoot, ecosystem and landscape scales in a boreal cutover in eastern Canada predominated by black spruce (Picea mariana Mill. BSP), lowbush blueberry (Vaccinium angustifolium Ait.) and sheep laurel (Kalmia angustifolia L.). We calculated CDD as the sum of minimum daily temperatures below a 5 degrees C threshold. Light-saturated photosynthesis at the shoot level (A(max)) of black spruce and V. angustifolium decreased steadily with increasing CDD once temperatures below the CDD threshold value became frequent in mid-September, whereas K. angustifolia showed a more irregular pattern. Tissue acclimation played an important role in the decrease in A(max) as the season progressed, but only V. angustifolium showed decreasing foliar nitrogen concentrations. Based on eddy covariance flux tower data, maximum daily gross primary productivity (GPP(max)-tower) at the ecosystem level was more strongly related to CDD (r(2) = 0.59) than was maximum daily net ecosystem exchange (r(2) = 0.32). The GPP(max) was likely influenced by both tissue acclimation and the direct effects of changing temperatures and irradiances on physiological rates. Mean daily GPP, calculated for consecutive 8-day periods for a 25 km(2) area around the tower by the MODIS MOD17A2 Collection 4 satellite algorithm (GPP- MODIS), decreased more rapidly with increasing CDD than did GPP(max)-tower. Although GPP-MODIS was closely correlated with mean daily GPP from the tower (GPP(daily)-tower, r(2) = 0.95) over the late growing season, the former was about twice as high. Although MODIS estimates of air temperature closely tracked the ground data, the maximum light-use efficiency parameter used by the MODIS algorithm was much higher than that indicated by the tower measurements. There was a 3% decline in GPP(max)-tower with an increase of 10 CDD, corresponding to the percent decline in branch-level A(max) of black spruce and V. angustifolium.  相似文献

During autumnal leaf senescence, leaf nitrogen in deciduous trees is translocated to storage sites, especially bark and xylem tissues. Proteins that accumulate in large amounts in bark and xylem in winter and are absent from these organs in summer are called storage proteins, and are believed to be vehicles for storing nitrogen reserves. These reserves are important for spring growth and help trees tolerate or recover from both abiotic and biotic stresses. Based on seasonal patterns of accumulation, we previously identified three storage proteins with molecular masses of 60, 19 and 16 kDa in bark tissues of 'Loring' peach (Prunus persica (L.) Batsch). To characterize the distribution of these proteins in different-aged tissues and to determine if they have any function other than nitrogen storage, we examined their seasonal distribution in bark tissues of current-year and 1-year-old shoots, scaffold branches, main trunks and 4-5-year-old roots of 'Loring' peach. Verification of protein identity was based on molecular mass and reactions with antibodies directed against each specific protein. Protein distribution was variable. For all three proteins, the greatest amount was present in mid-winter in current-year and 1-year-old shoots. These tissues also showed the greatest seasonal variation in the amount of protein present. The 16 kDa protein was present only in the youngest shoots, whereas the 19 kDa protein was present in all tissues examined. The 60 kDa protein was absent in root tissue. The amino acid composition and sequence of each protein were determined. The 60 kDa protein was identified as a dehydrin, and the 19 kDa protein appeared to be related to a family of allergen proteins in Rosaceous plants, some members of which are associated with pathogenesis-related proteins. The amino acid sequence of the 16 kDa protein appeared to have no homology with any proteins in the SwissProt database. Therefore, it is likely that the 16 kDa protein, in a strict sense, is a bark storage protein. Defining storage proteins solely by their pattern of accumulation and the extent to which they accumulate may not be a good functional definition. It is possible that storage proteins have functional roles in addition to nitrogen storage.  相似文献

Cutaneous haeangiomas are benign vascular neoplasms that arise from endothelial cells of blood vessels. Haemangiomas account for 0.6%–4% of all equine cutaneous neoplasms and the fetlock is the most commonly affected site. We describe the use of computed tomography angiography (CTA) for the evaluation of a cutaneous haemangioma located on the plantarolateral aspect of the left hindlimb fetlock of a 9-month-old Standardbred colt. Computed tomography angiography of the affected fetlock was performed under general anaesthesia. The medial plantar artery was catheterised and a total volume of 50 ml of iodinated non-ionic contrast medium (Iopamidol, 300 mg I/ml, Bracco Imaging Canada) was injected at a rate of 2 ml/s. Following contrast medium administration, the dorsal metatarsal artery and branches including the lateral and medial digital arteries were well demarcated. Two smaller lateral and medial arteries were also identified, forming the vascular network of the metatarsophalangeal joint. At the level of the haemangioma, two tortuous arteries arising from the lateral digital artery were identified, in addition to multiple small branches from nearby cutaneous arteries. These vessels supplied the homogeneously strongly contrast-enhancing cutaneous mass. The initial goal of the CTA study was to map the vascular anatomy for arterial embolisation in conjunction with pharmacological therapy. Considering the involvement of multiple small arterial branches, complete surgical excision along with ligation of the two main supplying arteries was alternatively elected, resulting in a successful long-term outcome.  相似文献

Intravenously administered docetaxel (DT) is problematic in cats because of the requirement for premedication to ameliorate acute vehicle-induced hypersensitivity reactions. Previously we have revealed that therapeutic plasma concentrations of DT can be achieved in normal and tumor-bearing dogs when DT is administered PO in combination with oral cyclosporin A (CSA). The purpose of this study was to identify the maximally tolerated dosage and characterize the pharmacokinetic disposition of oral DT combined with CSA in cats with tumors. Eighteen tumor-bearing cats were enrolled in this phase I dose escalation and pharmacokinetic study. DT was administered by gavage with CSA (5 mg/kg) twice over a 3-week period. The starting dose of DT was 1.0 mg/kg. Based on the clinical toxicity profile, with gastrointestinal adverse effects and hematologic toxicity the maximal tolerated dose of oral DT was 1.75 mg/kg in combination with 5 mg/kg CSA. Additional studies are necessary to determine the efficacy of DT/CSA in cats with epithelial tumors.  相似文献

Maternal plasma leptin is elevated in ewes during pregnancy. The authors studied whether there was any relation between maternal plasma leptin and insulin concentrations, the number of fetuses and the circulating and faecal levels of gestagens. At the end of the breeding season in January the ovarian activity of Prolific Merino ewes was induced/synchronised with gestagen + eCG treatment. Ewes were inseminated artificially (AI) by laparoscopy. Blood and faecal samples were collected before AI (day 0) and again 41, 81 and 101 days later. The plasma levels of leptin (pL), insulin and progesterone (pP4), and the faecal P4 metabolite (P4-met) content were determined. The day 0 level of pL was significantly higher in pregnant (n = 24) than in non-pregnant ewes (n = 32). By day 41 the pL of pregnant animals had doubled, it showed a further moderate increase on day 81, and decreased slightly thereafter. During pregnancy pP4 and faecal P4-met rose continuously and were positively correlated at all stages. The mean levels of pL and pP4 and the faecal content of P4-met were lower in ewes bearing single (n = 12) than in those with 2 (n = 6) or 3-5 fetuses (n = 6). Analysis of variance demonstrated significant differences according to the number of fetuses in the pL and pP4, but not in P4-met (p = 0.042, 0.044, and 0.051, respectively). Leptin showed positive correlation with insulin before the AI but not during pregnancy. On days 41 and 81 pL showed a slight positive correlation with P4 and P4-met, which decreased slightly by day 101. This study shows that although leptinaemia is affected by the number of fetuses and the level of P4, pregnancy stage is a more important regulator than these additional factors.  相似文献

Bovine viral diarrhoea virus (BVDV) is an endemic pathogen worldwide and eradication strategies focus on the identification and removal of persistently infected (PI) animals arising after in utero infection. Despite this, acute infections with BVDV can persist for months or years after the removal of the PI source despite repeated screening for PIs and tight biosecurity measures. Recent evidence for a prolonged duration of viraemia in the testicles of bulls following acute BVDV infection suggests the possibility of a form of chronic persistence that may more closely resemble the persistence strategies of hepatitis C virus (HCV). To investigate the potential for virus transmission from infected and recovered cattle to virus naïve hosts we established an acute infection of 5 BVDV-naïve calves and monitored animals over 129 days. Infectious BVDV was detected in white blood cells between days 3 and 7 post-challenge. The animals seroconverted by day 21 post-infection and subsequently were apparently immune and free from infectious virus and viral antigen.Animals were further monitored and purified white blood cells were stimulated in vitro with phytohaemagglutinin A (PHA) during which time BVDV RNA was detected intermittently.Ninety-eight days following challenge, blood was transferred from these apparently virus-free and actively immune animals to a further group of 5 BVDV-naïve calves and transmission of infection was achieved. This indicates that BVDV-infected, recovered and immune animals have the potential to remain infectious for BVDV-naïve cohorts for longer than previously demonstrated.  相似文献

OBJECTIVE: To evaluate gross, histopathologic, and serum biochemical findings caused by Leptospira interrogans serovars pomona and bratislava inoculated in dogs. ANIMALS: Twenty-seven 8-week-old female Beagles. PROCEDURE: Dogs were randomly assigned to challenge or control groups. Challenge groups were conjunctivally inoculated on 3 successive days with 5 x 10(7) L interrogans serovar pomona (n = 12) or serovar bratislava (11). Clinical signs were recorded throughout the experiment, and clinical pathology assays, bacteriologic culture, and necropsies (6 or 7 dogs necropsied at each time point) were done on postinoculation day (PID) 7, 10, 14, and 20. RESULTS: Infection could not be confirmed in any serovar bratislava-inoculated dog, and control dogs remained healthy throughout the experiment. Positive culture and fluorescent antibody test results were confirmed in 11 of 12 serovar pomona-inoculated dogs. Fever and lethargy starting at PID 7 were the most common clinical signs in serovar pomona-infected dogs. On day 10, gross lesions included multifocal renal and pulmonary hemorrhage and perirenal edema. Serovar pomona-inoculated dogs had histopathologic lesions including hepatitis, interstitial nephritis, and pneumonia at PID 7, 10, 14, and 20. Increases in BUN, anion gap, and bilirubin concentration occurred on PID 10, 14, and 20. Platelet counts in dogs with positive results of bacteriologic culture were decreased from baseline values on PID 10, 12, and 14. CONCLUSIONS AND CLINICAL RELEVANCE: Conjunctival inoculation with L interrogans serovar pomona resulted in a high rate of infection with concomitant hemorrhagic and inflammatory lesions of the kidneys, liver, and lungs.  相似文献
